چکیده
THE EFFECT OF EXPOSING RABBIT RETICULOCYTE RIBOSOMES TO CONCENTRATED SOLUTIONS OF POTASSIUM CHLORIDE WAS EXAMINED BY: (a) dialysis against 0.5m-potassium chloride; (b) zone centrifugation through a sucrose gradient in 0.5m-potassium chloride; (c) differential centrifugation of a solution made 0.5m with respect to potassium chloride. The products of each treatment and their ability to support protein synthesis in a reticulocyte cell-free system, in the presence and in the absence of polyuridylic acid, were examined. The following results were found. (1) Exposing the polysomes to 0.5m-potassium chloride was not a sufficient condition for the complete dissociation of ribosomes into subparticles; the reaction showed a concentration-dependence, implying the existence of an equilibrium between the various ribosomal species. Disturbance of the equilibrium by removing certain products, as in zone centrifuging, can lead to complete dissociation. (2) The subparticles produced by dialysis or sucrose-gradient fractionation were biologically inactive and unstable. (3) The pellet obtained by differential centrifuging consisted of subparticles, if suspended in a Mg(2+)-free buffer; addition of Mg(2+) converted about 30% of the material into heavier sedimenting species, and the preparation had 20-40% of the activity of the untreated control polysomes in the cell-free system. Addition of the 0.5m-potassium chloride supernatant fraction resulted in further apparent reconstitution of sub-particles into ribosomes and polysomes and in a 50-100% restoration of biological activity. When both polyuridylic acid and supernatant factors were present incorporations similar to or higher than those of the control were attained.

Reconstruction of the smaller subparticle of rabbit ribosomes from core-particle and split-protein fractions.
The smaller subparticle of rabbit reticulocyte ribosomes was shown to yield core-particle and split-protein fractions on treatment with 2.5 M-NH4Cl/61 mM-MgCl2. The core-particle fraction was inactive in poly(U)-directed polyphenylalanine synthesis, but activity was restored after recombination with the split-protein fraction. Optimum ionic conditions for the reconstruction of active subparticl...
متن کاملTranslation of exogenous messenger RNA for hemoglobin on reticulocyte and liver ribosomes.
The ribosome and initiation factor requirements for translation of rabbit-reticulocyte hemoglobin mRNA on rabbit reticulocyte ribosomes, reticulocyte ribosomal subunits, and liver ribosomes have been studied. Excellent synthesis of globin chains from exogenous mRNA in the fractionated cell-free system has been achieved. There is a near absolute requirement for each of the initiation factors, M(...
متن کاملReassembly of the peptidyltransferase centre of larger subparticles of rabbit reticulocyte ribosomes from a core-particle and split-protein fraction.
We report the reconstruction, from a core-particle and split-protein fraction, of the larger subribosomal particle of rabbit reticulocytes. The reassembled particle was active in polyphenylalanine synthesis and in the puromycin reaction. The core-particles and split-protein fractions were obtained by treatment of the larger subparticle with salt solutions containing NH4+ and Mg2+ in the molar r...
متن کاملProperties of membrane-bound ribosomes in reticulocytes.
An average of 17% of rabbit reticulocyte ribosomes are closely bound to the cell membrane. Evidence is presented to indicate that membrane binding of reticulocyte ribosomes is not an artifact. Membrane-bound reticulocyte ribosomes differ in physical and chemical properties from free reticulocyte ribosomes in having a high coefficient of sedimentation (greater than 400 S) and a high protein to R...
متن کاملInteraction of rabbit reticulocyte ribosomes with bacteriophage f1 mRNA and of Escherichia coli ribosomes with rabbit globin mRNA.
We have compared the behavior of a prokaryotic mRNA in a eukaryotic ribosome binding system and of a eukaryotic mRNA in a prokaryotic ribosome binding system. Using (32)P- and (125)I-labeled bacteriophage f1 mRNA, we have shown that rabbit reticulocyte 80S ribosomes can protect specific sequences from pancreatic RNase digestion, including those sequences protected by Escherichia coli ribosomes....
